Development of methods for preparation of monsubstituted derivatives

Our methods for preparation of monosubstituted derivatives are based on regioselective synthesis of allyl or cinnamyl CD derivatives [Jindrich1995], [Jindrich2005], [Rezanka2010].
These compounds can be used as precursors for relatively easy preparation or practically unlimited series of regioselectively substituted CD derivatives, which could be prepared by other means only with great difficulties.

Synthesis of C3 symmetric building units capable of self-assembly

C3 symmetric molecules with symmetrically 1,3,5-substituted benzen core, having the end groups containing cyclodextrin units or groups capable to form inclusion complexes with cyclodextrin, can be used for construction of self-assembled supramolecular structures, e.g. of the type of non-covalent dendrimers. Theoretically, they can also form molecular networks on suitable solid surfaces.

Preparation of chemosensors based on CD

If the fluorescent compound forms inclusion complex with cyclodextrin molecule the induced change of the fluorescence can be used for sensoric purposes.

If such fluorescent compound is covalently bound to CD, it forms the intramolecular complex. In the presence of other compounds (analytes) which can also form inclusion complexes with the CD the fluorescent group located in the CD cavity can be exchanged by the analyte and the measureable change of the fluorescence will appear.

Preparation of selectively multifunctionalized derivatives of CD to be used in MRI

Recently, the cyclodextrin derivatives were proved to be highly effective contrast agents in magnetic resonance imaging (MRI) in medicine. The fact that CD contains large number of symmetrically aranged hydroxy groups located very close to each other was utilized in this case. On these hydroxy groups can be covalently bound gadolinium complexes which are widely used in MRI. Besides these molecules in which the Gd complexes are bound only to one side of CD molecule, also other type of compounds can be prepared, where on the other side of the CD are connected other "targeting" groups which can deliver the whole molecule to the desired tissue where the targeting group is selectively bound.

Study of reactions in microreactor in overheated water solution

Na našem spolupracujícím pracovišti, kde byla vyvinuta před lety technologie Lab-on-chip, byl nedávno vyvinut nový typ mikroreaktoru, ve kterém lze zahřívat vodu za atmosferického tlaku až na 220 °C bez toho, aby začala vřít. Tak se otevřela nová cesta pro zkoumání známých hydrolytických reakcí ale za dosud naprosto neznámých podmínek. Předmětem projetku je tedy studium kinetiky, analyza produktů a zjištování selektivity hydrolyzačních reakcí.
